dc.description.abstractBu çalışmada, albino Wistar erkek sıçanlara subkronik uygulanan alpha-cypermethrinin testis, epididimis, prostat ve seminal vezikül dokularındaki etkilerinin histopatolojik ve immünohistokimyasal olarak değerlendirilmesi amaçlanmıştır. Çalışmada 2 kontrol ve 3 doz grubu oluşturulmuştur. Alpha-cypermethrin sıçanlara oral yolla mısır yağında çözülerek verilmiştir. Sıçanların dokularından alınan kesitler hem rutin histolojik ve indirekt immünohistokimyasal yöntemle boyanmış ve sonuçlar ışık mikroskobu kullanılarak değerlendirilmiştir. Histopatolojik incelemeler neticesinde testiste sertoli hücrelerinde vakuolizasyon, tübül dejenerasyonu, lümende spermatojenik hücre ve ödem; epididimis lümeninde spermatojenik hücreler, sperm içermeyen tübüller ve bağ dokuda artış; prostat dokusunda lümende daralma, salgıda çekilme, mononükleer hücre infiltrasyonu ve epitelial katlanma; seminal vezikülde salgıda azalma, epitel hücrelerinde mitoz ve epitelial katlanma tespit edilmiştir. İmmünohistokimyasal boyanmada dozlara bağlı olarak epidermal büyüme faktörü reseptörü (EGFR) dağılımı açısından farklılıklar mevcuttur. En şiddetli boyanma epididimis ve seminal vezikül epitelinde gözlenmiştir. Boyanma, diğer dokularda daha zayıftır. Histopatolojik ve immünohistokimyasal bulgular birlikte değerlendirildiğinde alpha-cypermethrinin ADI (0-0,02mg/kg) değerine yakın doz grubunda (0,09mg/kg/gün) da erkek üreme sisteminde etkili olabildiği ve doz artışıyla birlikte bulguların şiddetinde artışa neden olduğu sonucuna varılmıştır. dc.description.abstractThe aim of this study was to assess histopathologically and immunohistochemically the effects of subchronicly administered alpha-cypermethrin on testis, epididymis, prostate and seminal vesicle in male albino Wistar rats. Two control groups and three experimental groups were established. Alpha-cypermethrin was given to rats orally by dissolving it in corn oil. Sections from the tissues were stained for both routine histological methods and for indirect immunohistochemical methods, and then results were evaluated under light microscope. Histopathological assessments revealed vacuolization in Sertoli cells, tubule degenaration and spermatogenic cells and edema within the lumen; spermatogenic cells in epididiymic lumen, tubules without sperm cells and increase in connective tissue; narrowing of the lumen of prostate tissue, lack of secretion, mononucleer cell infiltration and epithelial folding; lack of secretion in seminal vesicle tissues, mitosis of epithelial cells and epithelial infolding. In immunohistochemical staining there was dose dependent difference regarding epithelial growth factor receptor (EGFR) distribution. The most intense staining was observed in epididymis and vesicula seminalis epithelium. Staining is less intense in other tissues. When histopathological and immunohistochemical findings were evaluated together, it was concluded that alpha-cypermethrin could be effective in male reproductive system in the dose group (0.09 mg/kg/day) which is near to the ADI (0-0.02 mg/kg) value and increase in intensity of the results was observed along with the increasing dose.
