dc.description.abstractBu çalışmada Çanakkale ili ve ilçelerinde 2011-2012 üretim yılları içinde bir sörvey çalışması yürütülerek Elma mozaik virüsü (Apple mosaic virus; ApMV)?ne benzer simptom gösteren elma ağaçlarından 44 örnek toplanmıştır. Toplanan örnekler ApMV?nin varlığının belirlenmesi amacıyla DAS-ELISA yöntemiyle test edilmiştir. DAS-ELISA testi sonucunda 44 örnekten 28?i (% 63) ApMV ile infekteli bulunmuştur. İnfekteli bulunan örnekler içerisinden seçilen 8 izolat, RT-PCR ürünleri kullanılarak Tek Sarmal Konformasyonel Polimorfizm (Single-stranded Conformation Polymorprhism; SSCP) yöntemi ile analiz edilmiş ve izolatlar arasında farklılık olmadığı belirlenmiştir. DAS-ELISA ve RT-PCR analizleri sonucu pozitif olduğu bilinen bazı izolatlara yapılan Western blot analizi sonucunda 25 kDa ağırlığında proteinler elde edilmiştir. dc.description.abstractIn this study, a survey was conducted in Çanakkale Province and sub provinces on apple trees that show Apple mosaic virus-like symptoms between the years 2011-2012. A total of 44 samples were collected from these areas and tested by DAS-ELISA for the presence of ApMV. As a result of the DAS-ELISA analysis 44 out of 28 samples were found infected with ApMV. Eight isolates selected from the infected isolates were analyzed with RT-PCR products by using Single-stranded Conformation Polymorprhism (SSCP) method and the no difference was found among the isolates. As a result of Western blot analysis applied to positive isolates determined by the result of DAS-ELISA and RT-PCR analysis, 25 kDa weight proteins were obtained.
