dc.description.abstractBu çalışmada, Çanakkale ili Merkez ilçeye bağlı Kepez ve Güzelyalı beldeleri ile Karacaören köyünde soğanlı süs bitkilerinin yetiştiriciliğinin yapıldığı ev bahçelerinde 2012-2013 yıllarında bir sörvey çalışması yürütülerek Hıyar mozaik virüsü (Cucumber mosaic virus; CMV)'ne benzer simptom gösteren nergis, sümbül, zambak gibi soğanlı süs bitkilerinden 67 örnek toplanmıştır. Toplanan örnekler, CMV'nin varlığının belirlenmesi amacıyla DAS-ELISA yöntemiyle test edilmiştir. DAS-ELISA testleri sonucunda 67 örnekten 12'si (%18) CMV ile infekteli bulunmuştur. Belirgin olarak virüs-benzeri simptom göstermesine karşın DAS-ELISA testi sonucunda şüpheli bulunan örnekler ile negatif olarak tespit edilen örneklerden rastgele seçilen iki örnek RT-PCR analizi ile test edilmiştir. RT-PCR testleri sonucunda da bu örnekler negatif olarak bulunmuştur. DAS-ELISA testleri sonucunda pozitif olduğu bilinen bazı izolatlarla yapılan Western blot analizi sonucunda 24 kDa moleküler ağırlığa sahip bantlar elde edilmiştir. dc.description.abstractIn this study, a survey was conducted in home gardens of Kepez and Güzelyalı towns and Karacaören village connected to the central district of Çanakkale Province in ornamental bulb plants like daffodils, hyacinths and lilies that show Cucumber mosaic virus-like symptoms in year 2012-2013. A total of 67 samples were collected from these areas and tested by DAS-ELISA for the presence of CMV. As a result of the DAS-ELISA (18%) 12 out of 67 samples were found infected with CMV. Two randomly selected samples from the samples that show significant symptoms with negative results from DAS-ELISA test and samples that resulted suspicious in the test, are tested with RT-PCR analysis. These samples were found to be negative as the result of RT-PCR. As the result of Western blot analysis with positive isolates from DAS-ELISA tests, bands with 24 kDa molecular weight were obtained.
