Özet
Bu araştırmada, Çanakkale yöresinde ve Kazdağı florasında yetişen bazı tıbbi ve aromatik bitkilerin doku kültürüne yanıtlarının belirlenmesi amaçlanmıştır. Araştırma Çanakkale Onsekiz Mart Üniversitesi Ziraat Fakültesi Tarla Bitkileri Bölümü'nde yürütülmüştür. Lamiaceae familyasındaki Melissa, Oreganum, Sideritis, Salvia, ve Thymus Kazdağı florasında bulunan bazı tıbbi ve aromatik bitki türleridir. Denemelerde Murashige ve Skoog (MS) temel besi ortamı kullanılmış ve bu bitki türleri üzerine farklı bitki büyüme düzenleyicilerin etkileri incelenmiştir. Kekik, oreganum ve oğulotu bitkisinin yetiştiği en uygun ortamın büyüme düzenleyici içermeyen MS besi ortamı olduğu bulunmuştur. Adaçayı bitkisinde koyu yeşil ve yumuşak yapıdaki kallus gelişimi MS+0,5mg/l BA+0,5mg/l NAA'lı ortamda, kahverengi sert yapıdaki kalluslar ise 0,5mg/l BA+2,0mg/l NAA besi ortamından elde edilmiştir. Sonuç olarak bu araştırmada incelenen bitki türleri (Sideritis trojana hariç) doku kültürüne başarı ile aktarılmış ve çoğaltılmıştır.
In this study, the aim was to determine tissue culture responses of some medicinal and aromatic plants grown in flora of Ida Mount and Çanakkale Region. The study was carried out at Department of Field Crops, Agricultural Faculty, Canakkale Onsekiz Mart University. Lemon balm, oreganum, siderites, sage and thyme which belong to Lamiaceae family were investigated. In the experiments, Murashige and Skoog (MS) was used as basal medium and the effects of different growth regulators on growth of these species were examined. The results showed that the best medium for lemon balm, oreganum and thyme was to be MS medium without growth regulators. In sage, dark green and soft callus growth was obtained on MS medium supplemented with 0.5mg/l BA and 0.5mg/l NAA whereas brown and hard calli were obtained on MS medium supplemented with 0.5mg/l and 2.0mg/l NAA. As a result, all plant species (except Sideritis trojana) investigated in this study were successfully transferred to tissue culture and micropropagate.
