Öğe Arabidopsis thaliana (L) Heynh. türünde ß-1,3-glukanaz kodlayan bazı genlerin fonksiyonlarının incelenmesi(Çanakkale Onsekiz Mart Üniversitesi, 2012) Polat, Elif; Taşkın, Kemal MelikBu çalışmada Arabidopsis thaliana (L.) Heynh. anterlerinde ß-1,3 glukanaz (kallaz) kodlayan AT3G23770 ve AT4G14080 genlerinin kalloz yıkımındaki fonksiyonunun belirlenmesi amaçlanmıştır. Bu amaçla, bu genlerin T-DNA nakavt mutantlarına ait tohumlar ABRC Stok Center (Columbus)'dan Prof. Dr. Rod J. Scott (Bath üniversitesi, İngiltere) aracılığı ile temin edilmiştir. Çalışmada, AT3G23770 geni için N859965 stok numaralı hat, AT4G14080 geni için ise CS850174 stok numaralı hat kullanılmıştır. Tohumlar labarotuvar ortamında çimlendirilmiş ve her iki mutant hata ait bitkiler fenotipik olarak incelenmiştir. Buna göre bitkiler 3-4 hafta sonrasında çiçeklenmeye başlamış ve baklalar gelişmiştir. Bu bitkilerin genotipleri gen spesifik PCR ile belirlenmiştir. PCR sonuçlarına göre N859965 mutant hattı için homozigot (aa), CS850174 hattı içinse heterozigot (Bb) bireyler belirlenmiştir. Ayrıca, bu bitkilerde erkek kısırlık ve bakla gelişimi incelenmiştir. Daha sonra, çalışmada her iki mutasyonu bir arada taşıyan hatlar (double mutant) elde etmek için melezlemeler yapılmıştır. Buradan elde edilen tohumlardan çimlenen melezlerin genotipleri özgün PCR aracılığı ile belirlenip homozigot genotipe sahip (aabb) bitkiler seçilmiştir. Double mutant bitkilerde çiçek, polen ve bakla gelişimleri hem sitolojik hem de fenotipik olarak incelenmiş, bakla başına düşen tohum miktarı belirlenmiştir. Çalışmada ayrıca, ilgili genlerin anlatımları RT-PCR ile araştırılmıştır.Öğe Evaluation of The Frequency of Artifacts in CBCT Depending on The Different Size of Field of View(Marmara University, 2023) Polat, Elif; İncebeyaz, Burak; Deniz, Hatice Ahsen; Eren, HakanABSTRACT Objectives: Various artifacts can be encountered when examining Cone-beam computed tomography(CBCT) images. In recent years, no descriptive study has been conducted on CBCT artifacts in the literature. The aim of this study is to identify artifacts in CBCT images with different imaging fields (FOV) taken on the same device, to determine their frequency, and also to reveal artifacts that are little known in the literature. Materials and Methods: In the study, CBCT images of the Faculty of Dentistry between the years 2012-2021 were scanned retrospectively and a total of 359 Cone Beam Computed Tomography images with 50x55, 100x55, 100x90, 130x55, 130x90, 230x170, 230x270 imaging fields (FOV) in the database, which met the exclusion and inclusion criteria, were analyzed by three oral and maxillofacial radiology research assistants and the types of artifacts seen on these images were determined. The incidence of the identified artifact types in the specified imaging areas was evaluated. Results: When looking at all images, the most common errors, regardless of FOV, were inevitable artifacts. Aliasing and motion artifacts were seen at higher rates on CBCT images with a larger field of view. In addition, the ring artifact was encountered in CBCT images with high imaging fields such as 130x90, 230x170 and 230x270. The case is the first reported case of OSCC after orthodontic treatment in a low-risk young female patient.Öğe Royal jelly attenuates gastric mucosal injury in a rat ethanol-induced gastric injury model(Springer, 2020) Duran, Yasin; Karaboga, Ihsan; Polat, Fatin Rustu; Polat, Elif; Erboga, Zeynep Fidanol; Ovali, Mehmet Akif; Oztopuz, Rahime OzlemThe aim of the study was to investigate traditionally used Royal Jelly (RJ) for treating an ethanol-induced gastric ulcer model in rats. A total of 32 Wistar albino male rats were divided into 4 groups of 8: group I = Control, group II = Ethanol, group III = RJ + Ethanol, and group IV = Lansoprazole + Ethanol. In groups II, III, and IV, animals were administered 1 ml of absolute ethanol orally after a 24-h fast to induce ulcer formation. The histopathological changes in the gastric mucosa were determined using hematoxylin-eosin (H&E) staining. Immunohistochemically, inducible nitric oxide (iNOS) and nuclear factor kappa beta (Nf-kappa beta) markings were evaluated in gastric tissue. Cell death in the gastric mucosa was determined by the TUNEL method. Oxidative status markers, superoxide dismutase (SOD), malondialdehyde (MDA), catalase (CAT), and myeloperoxidase (MPO) levels were determined spectrophotometrically. Expression of the interleukin - 1 beta (IL-1 beta) and tumor necrosis factor-alpha (TNF-alpha) genes in gastric tissues was determined by real-time PCR; and TNF-alpha, IL-10, and IL-1 beta levels were determined. RJ was found to inhibit iNOS and Nf-kappa beta activity in the gastric mucosa and prevent epithelial cell apoptosis. In particular, pro-inflammatory cytokines TNF-alpha and IL-1 beta levels were significantly decreased in the RJ + Ethanol group compared to the Ethanol group. In addition, a decrease in the MPO level indicated that RJ prevented tissue damage, especially by preventing inflammatory cell infiltration.
