    Detection of Citrus tristeza virus (CTV) from Satsuma Owari mandarins (Citris unshiu) by direct tissue blot immunoassay (DTBIA), DAS-ELISA, and biological indexing

    Three different citrus-producing regions of Turkey-Edremit Gulf, Coastal Aegean, and eastern Black Sea-were surveyed in 2005 and 2006. A total of 119 samples were collected from Satsuma Owari mandarin (Citrus unshiu) trees grafted on Citrus tristeza virus (CTV)-resistant Poncirus trifoliata rootstocks in commercial groves and home gardens. All samples were tested for the presence of CTV by direct tissue blot immunoassay (DTBIA) and double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) using young shoots. The samples that tested positive were indexed for biological properties. The results obtained from DTBIA tests showed that 20 of 119 (16.8%) tested trees were infected with CTV and 99 trees were virus free. All samples that tested positive in DTBIA were also positive in DAS-ELISA. After biological indexing, none of these 20 isolates showed any symptoms on sour orange, grapefruit, and sweet orange plants, but all isolates induced vein-clearing symptoms on Mexican lime (Citrus aurantifoli) in 9 months. Some isolates also caused leaf-cupping and chlorosis in Mexican lime. However, 15 months after initial grafting, all isolates induced mild to moderate stem-pitting on Mexican lime. In addition, young tissues, petioles, and leaf samples were collected periodically at monthly intervals for I year (2006) from a 25-year-old Satsuma Owari mandarin grafted on P. trifoliata in the Edremit Gulf. CTV was readily detected in tissue blots of young shoots and petioles from CTV-infected plants during the autumn, winter, and spring seasons. Similarly, the highest ELISA values were obtained in April; the lowest values were noted in September. This study showed that DTBIA is a rapid, sensitive, and reliable procedure for detecting CTV under field conditions.

    Determination of Virus and Viroid Diseases on Satsuma Owari Mandarins by Biological and Serological Methods in Edremit Gulf Region

    This study was carried out during 2005-2006 in citrus orchards in Edremit Gulf Region (Edremit, Havran and Burhaniye subprovince) to determine prevalence of virus and viroid diseases including Exocortis, Citrus tristeza virus, Satsuma dwarf and Psorosis on Satsuma Owari mandarins. During 2005-2006 in citrus production areas in Edremit Gulf region were surveyed and samples were collected from symptomatic trees. Samples were tested by biological indexing for Exocortis and Satsuma dwarf, DAS-ELISA for Psorosis and DAS-ELISA and biological indexing for Citrus tristeza virus. DAS-ELISA test reveal that while 38 of 156 samples were infected with Citrus tristeza virus, none of the 10 samples tested was positive for Psorosis. Biological indexing showed that 1 of 20 samples tested was infected with Exocortis viroid, however no symptoms were observed from the Satsuma dwarf samples. On the other hand, indexed Citrus tristeza virus isolates induced vein clearing and stem pitting symptoms on Mexican lime plants.

    Edremit Körfez bölgesi'ndeki satsuma owari mandarinlerinde yaygın olan virüs ve viroid hastalıklarının biyolojik ve serolojik yöntemlerle saptanması

    Bu çalışma, Edremit Körfez Bölgesi’nde yer alan Edremit, Havran ve Burhaniye ilçelerindeki turunçgil bahçelerinde 2005 ve 2006 yılları arasında Satsuma Owari mandarinlerinde virüs ve viroid hastalık etmenlerinden Exocortis, Tristeza, Satsuma dwarf ve Psorosis’in varlıklarının belirlenmesi amacıyla yürütülmüştür. Bu amaçla Edremit Körfez Bölgesi turunçgil üretim alanlarında bir survey çalışması yapılarak belirtilen hastalık etmenlerinin oluşturduğu simptomlara benzer belirtiler oluşturan ağaçlardan örnekler alınmış ve alınan örnekler Exocortis viroidi ve Satsuma dwarf virüs hastalıkları için biyolojik indeksleme, Psorosis için DAS-ELISA ve Tristeza içinse hem DAS-ELISA hem de biyolojik indeksleme yöntemleri uygulanarak test edilmiştir. Yapılan DAS-ELISA testi sonucunda Tristeza için alınan 156 örnekten 38’i pozitif bulunurken, Psorosis için alınan 10 örnekten hiçbirinde pozitif sonuç elde edilememiştir. Yapılan biyolojik indeksleme çalışmalarında ise Exocortis viroidi için alınan 20 örnekten 1’inde hastalık simptomlarına rastlanılırken, Satsuma dwarf için indekslenen 15 örnekte ise herhangi bir simptom gözlenmemiştir. Ayrıca DAS-ELISA testi sonucunda Tristeza için pozitif bulunan örneklerden 6’sı biyolojik indekslemede kullanılmış ve indikatör bitkilerden Meksika laymında damar açılması ve gövde çukurlaşması simptomları gözlenmiştir.

    Occurrence and molecular characterization of Turkish isolates of Turnip mosaic virus

    A total of 142 samples of plants showing symptoms of Turnip mosaic virus (TuMV) were collected from fields planted to Brassicaceae and non-Brassicaceae crops in the southwest Marmora region of Turkey, during the 2004-06 growing seasons. Using enzyme-linked immunosorbent assay (ELISA) TuMV was detected in the main brassica-crop fields of Turkey, with an overall incidence of 13.4%. TuMV was detected in samples from Brussels sprouts, cabbage, wild mustard, radish and wild radish, but not cauliflower or broccoli. The full-length sequences of the genomic RNAs of two biologically distinct isolates, TUR1 and TUR9, were determined. Recombination analyses showed that TUR1 was an intralineage recombinant, whereas TUR9 was a non-recombinant. Phylogenetic analyses of the Turkish isolates with those from the rest of the world showed that the TUR1 and TUR9 isolates belonged to world-Brassica and Asian-Brassica/Raphanus groups, respectively. This study showed that TuMV is widely distributed in the Asia Minor region of Turkey.
