    Development of melatonin-embedded PLGA-PEG6000 nanofiber biomaterial, and investigation of the effects on abdominal adhesion formation

    Abdominal adhesions are still among the most common postsurgical peritoneal inflammation-related complications. Adhesion-related disorders are still highly costly and prevalent due to advances in surgical techniques, treatment methods, and various drugs. The present study aimed to investigate the effects of Poly (D,L-lactide-co-glycolide) (PLGA)- polyethylene glycol (PEG6000) Nanofiber + Melatonin on the abdominal adhesion model in rats. For this purpose, PLGA-PEG6000 Nanofiber + Melatonin matrix was fabricated and implanted in an experimental abdominal adhesion model in rats. Our study consisted of an in vitro and an in vivo part. The degradation and release profile of the matrix and Melatonin (Mel) embedded matrix was performed in vitro. In vivo, the procedure was carried out with 18 Wistar male rats. Rats were divided into three groups as follows: Sham, Matrix, and Mel + Matrix, respectively. Consequent to degradation and release profiling in vitro, an experimental adhesion model was created and fabricated pure matrix (2 × 2 cm2), and matrix (2 × 2 cm2, 0.25 mg melatonin/per matrix embedded) was applied to injury area in related groups. Intra-abdominal adhesion scores were determined on post-op 21st day, under general anesthesia. Following, cecum, peritoneal tissue, and adhesive bands were harvested. Macroscopic analysis (severity of adhesion formation), Hematoxlyn&Eosin and Masson’s Trichrome staining (for the examination of the levels of infiltration of inflammatory cells, fibrosis, and neovascularization) were performed for the evaluation of the effects of Mel embedded and pure matrix Our results indicated that PLGA-PEG6000 Nanofiber + Melatonin matrix was degraded completely in rats abdominal cavity and significantly reduced adhesion formation compared to other groups macroscopically (p < 0.05). On the other hand, the histopathological analysis indicated that the fabricated matrix reduced inflammatory cell infiltration, fibrosis, and neovascularization levels.

    Investigation of Protective Effect of Hypericum Perforatum on Ethanol-induced Acute Gastric Mucosal Injury in Rats

    Objective: This study was conducted to evaluate the antiapoptototic and antiinflammatory effect of Hypericum perforatum in the rats induced gastric ulcer by absolute ethanol. Materials and Methods: Forty male Wistar albino rats were used in this study. Rats were divided four group, randomly. Group I (Control, n=10), Group II (Ethanol, n=10, 1 mL ethanol, orally, 90 min)  Group III (H. perforatum + ethanol, 1 mL H. perforatum, 120 min; 1 mL ethanol, 90 min, orally), Group IV (Olive oil + ethanol, 1 mL olive oil, 120 min; 1 mL ethanol, 90 min, orally). Gastric ulcer score  was determined in gastric mucosa by morphometrically. Histopatological staining, Immunohistochemical iNOS and PCNA staining, apoptotic TUNEL staining, gastric IL-10 and IL-6 expression (ELISA) and SOD, MDA, CAT levels were assessed in gastric tissue. Results: The results showed that ethanol incuded gastric damage, higher gastric ulcer index score, increased proinflammatory IL-6 level, elevated number of iNOS and TUNEL-positive stained cell number as well as higher MDA level in the group II (ethanol). Pre-treatment of H. perforatum and olive oil (group III and IV) significantly attenuated the gastric lesions as compared to the group II (ethanol). Conclusion: It was concluded that H. perforatum and olive oil may represents a potential therapeutic option to reduce the risk of gastric mucosal ulceration

    The effect of ozone (O3) therapy in addition to high dose methylprednisolone on hypoxia inducible factor-1 alpha (HIF-1?) expression in rabbit cornea

    Introduction: To investigate the effect of ozone (O3) therapy on cornea subjected to systemically used high dose methylprednisolone (MP) ina rabbit model.Methods: Twenty-four New Zealand White adult male rabbits were randomly divided into three equal groups as containing eight animals. Thefirst group (n = 8) was used as the control group and nothing was applied to them, whereas the other 2 groups named as steroid groups weresubjected to IM methylprednisolone injection at a dose of 20 mg/kg/day for three days. After three days of MP administration, only the third groupwas treated with 50-µg/mL O3 (20 mL O3) through the rectal insufflation for 14 sessions. The histopathological examination of corneas of threegroups were made, and they were also assessed regarding the expression of hypoxia-inducible factor-1 ? (HIF-1?).Results: It was determined that systemically administered high dose MP caused erosion and necrosis in corneal epithelium and stromaldisintegrations in corneal stroma in steroid groups (Group 2 and Group 3). In the MP + O3 group (Group 3), the histopathological findings weremild. The expression of HIF1-? in the cornea of Group1 (control group), Group 2 (MP), and Group 3 (MP-O3) was measured as, 17.9±9.6%,3.1±1.0% and 6.4±1.9% respectively.Conclusions: MP and MP-O3 therapy decreased HIF-1a expression in rabbit cornea in both intervention groups. Between these two groups,HIF-1? expression remained relatively high in the MP-O3 group than in the MP group alone.
