    Blood Cell Types and Abnormalities in Free Ranging Hermann’s Tortoise (Testudo hermanni) Populations from Different Habitats

    Testudo hermanni is considered near threatened globally in the wild. In this study differential leukocyte percentages, nuclear abnormalities, and erythrocyte morphology abnormalities of Testudo hermanni populations from two different habitat types in the Thrace Region were determined to whether habitats have a potential effect on blood cells. A total of 37 tortoises, 18 from an open habitat in Kırklareli/Karakoç, and 19 from an isolated habitat in Edirne/Keşan were studied. According to the results obtained from the Micronucleus Test, which detects genotoxic damage, there was no statistically significant difference between the two habitats. There were statistically significant differences in erythrocyte morphology abnormalities and differential leukocyte count between two populations, such as frequencies of anucleated erythrocyte, nuclear shift, elliptical shape distortion, monocyte and eosinophile percentages. This is the first study examined nuclear abnormalities of Testudo hermanni in Türkiye.

    Erythrocytic and Nuclear Abnormalities in Natrix sp. from the Biga Stream (Çanakkale, Türkiye)

    The Grass Snake (Natrix natrix) and the Dice Snake (Natrix tessellata) are potential bioindicator water snake species in certain aquatic habitats. The aim of this study is to evaluate the frequency of erythrocytic abnormalities, immature erythrocyte index, and nuclear abnormalities in Natrix natrix and Natrix tessellata from Biga Stream in Çanakkale, Türkiye. Erythrocytic abnormalities and nuclear abnormalities such as micronuclei, lobbed nuclei, notched nuclei, blebbed nuclei, and binucleate cells were observed in peripheral blood smears for each sample. According to our results, total erythrocytic abnormalities in N. natrix (3.01±0.55%) were higher than in N. tessellata (2.52±0.32%). Similarly, the frequency of total nuclear abnormalities in N. natrix (13.40±2.7%) was higher than in N. tessellata (12.68±5.81%) but there was no statistically significant difference in total erythrocytic and nuclear abnormalities. However, student's t-test revealed that there was a statistically significant difference between the two species only in terms of immature erythrocyte index (p: 0.008) which was higher in N. natrix. Due to the results, it was suggested that N. natrix may exhibit more physiological responses to envirenmental stress factors. © 2024, ABADER (Adıyaman Bilimsel Arastırmalar Dernegi).     Haematological and Genotoxicological Effects of Water Quality on Pelophylax bedriagae (Camerano, 1882) (Anura: Ranidae) and Mauremys rivulata (Valenciennes, 1833) (Testudines: Geoemydidae) in Çanakkale, Türkiye

    Haematological and genotoxicological parameters of the Levant green frog Pelophylax bedriagae (Camerano, 1882) and the Western Caspian turtle Mauremys rivulata (Valenciennes, 1833) were studied. These species inhabited two sites with differing water quality in the Sarıçay Stream passing through the city centre of Çanakkale, Türkiye. We examined whether the two species were affected by the water quality classes identified based on physical and chemical properties of the water. Statistically significant differences were found between the two sites in terms of the frequencies of micronucleus, lobbed, notched, kidney-shaped and blebbed nuclei, as well as total nuclear abnormality in Pelophylax bedriagae. Moreover, a higher rate of nuclear abnormalities was observed at the more polluted site, classified as water quality class IV. Statistically significant differences were observed in the haematocrit value, percentages of lymphocytes, eosinophils, heterophils and basophils, erythrocyte width and frequencies of lobbed and kidney–shaped nuclei as well as the heterophil/lymphocyte ratio, often used as a stress indicator, in Mauremys rivulata. In this species, it was determined that parameters other than lymphocyte and eosinophil percentages were higher in specimens from the more polluted site. © 2024 Institute of Biodiversity and Ecosystem Research - Bulgarian Academy of Sciences.     Herpetofaunal Diversity of Çanakkale Southwest Coastal Zones

    Along with the literature information obtained from previous studies, the determination of species in herpetofauna studies gives information about the herpetofauna of the research area. Researching the herpetofauna of regions is very important in terms of conservation of species, revealing biodiversity, identifying possible threats, and determining the preventitive measures to be taken against these threats. The study area is the southwestern coastal regions of Çanakkale, which is also the westernmost coast of Anatolia. This area consists of the localities of Ahmetçe, Sazlı, Kozlu, Behram, Bektaş, Koyunevi, Babakale, Gülpınar, Tuzla, Kösedere, and Tavaklı. Because it has the potential to be a coastline separated by the end of the Kaz Mountains, this study area has different habitats and has the potential to host species that exceed isolation of the Kaz Mountains. In this study, the amphibian and reptile diversity of terrestrial and aquatic areas along the coast of Southwest Anatolia starting from the end of the Kaz Mountains, which is the habitat preferences of the species, and the effects of environmental and anthropogenic factors on the herpetofauna of the region were investigated. A total of 5 amphibian species and 16 reptile species have been identified in the southwestern coastal areas of Çanakkale, and the Testudo graeca species are vulnerable according to the IUCN categories.

    Sarıçay (Çanakkale)'daki çevresel parametrelerin Pelophylax bedriagae ve Mauremys rivulata türleri üzerinde hematolojik ve genotoksikolojik etkilerinin belirlenmesi

    Bu çalışmada, Sarıçay'ın farklı su kalitelerine sahip olan iki lokalitesinde yaşayan Pelophylax bedriagae (Levanten Ova Kurbağası) (Camerano, 1882) (12♂♂, 8♀♀), Mauremys rivulata (Balkan Çizgili Kaplumbağası) (Valenciennes, 1833) (4♂♂, 17♀♀) türlerine ait toplam 41 adet örneğin, yaşam ortamlarındaki bazı ekolojik parametrelerden etkilenip etkilenmediği hematolojik ve genotoksikolojik analizler yapılarak belirlenmiştir. Klinik hematolojiye ait eritrosit sayısı, eritrometrik ölçümler, lökosit sayısı, lökosit tipleri (lökosit formülü), hemoglobin değeri, hematokrit değeri, ortalama eritrosit hacmi (OEH), ortalama eritrosit hemoglobini (OEHb), ortalama eritrosit hemoglobin konsantrasyonu (OEHbK); genotoksikolojik analizler kapsamında nüklear anormallik frekansları ve kandaki ağır metal birikimleri incelenmiştir. Analizler için kurbağaların orta abdominal veninden, kaplumbağaların ise dorsal kaudal veninden alınan kan örneklerinin bir kısmı eser element tüplerine aktarıldıktan sonra santrifüj edilerek elde edilen plazma kısmı ağır metal analizine gönderilmiştir. Diğer hematolojik parametreler manuel olarak veya preparatlar üzerinde gerekli incelemeler yapılarak gerçekleştirilmiştir. Sonuç olarak; yaşadıkları habitatların su kalitesine göre P. bedriagae türünde mikronükleus, loblu nükleus, çentikli nükleus, böbrek şekilli nükleus, tomurcuklu nükleus ve total nüklear anormallik frekanslarında; M. rivulata'da ise hematokrit değerinde, lenfosit, eozinofil, heterofil ve bazofil yüzdelerinde, eritrosit genişliği değerinde, loblu ve böbrek şekilli nükleus frekanslarında istatistiksel olarak anlamlı farklılık olduğu tespit edilmiştir.
